Analysis of AGE-RAGE interaction and cytokine expression in the periodontium of diabetic rats

Periodontal diseases (DPs) are chronic inflammatory diseases of supporting structures of the theet. Among diverse risk factors, diabetes exerts notable influence on periodontium. The presence of diabetes result in more severity and prevalence of DPs in human and experimental models. In diabetic rats, periodontal disease induced by ligatures and/or bacterial inoculation is more severe. However, in the absence of aggressive stimuli as ligatures, the influence of diabetes on periodontium of rat is poorly known. Our previous results show that only diabetes induction increase severity and prevalence of periodontal disease in rats. It was observed reduction in collagen fibers, fibroblasts, blood vessels and increase in the number of osteoclasts and inflammatory cells. In this way, diabetes is responsible for presence and progression of periodontal disease in rats characterized for alveolar bone loss and destruction of conjunctive periodontal tissue. In this project, we will evaluate the expression of inflammatory citokynes and AGE-RAGE interaction. It is known that expression of inflammatory citokynes and interaction AGE-RAGE are involved in pathogenesis of periodontal disease, however the mechanisms is poorly known. Initially the induction of diabetes in Wistar rats will be through (n=25) for alloxan administration (42 mg/kg) and analyze after 1, 3, 6, 9 and 12 months of diabetes induction, as previously demonstrated in our studies. It will be analyze by imunohistochemical the expression of TNF-, IL-1, MMP-2, MMP-9 and RANKL. Then, it will be evaluated the AGE-RAGE interaction with receptors RAGE. This results will be evaluated with our previous results related to relation between diabetes and periodontal disease. These data will contribute to the knowledge of role of diabetes in the pathogenesis of periodontal diseases, and may provide the basis for future therapeutic interventions in the inflammatory process of these pathologies.
