Abstract

The objective of this research ex vivo is to evaluate in stereomicroscope and cofocal microscopy, the quality of the filling provided by the technique of the vertical condensation, the single cone, RealSeal and GuttaFlow through the use of cones of gutta-percha and Resilon with conicity varied in accordance with different rotary systems in the instrumentation: ProTaper and K3 endo systems. Two hundred mesial roots of inferior human molars obtained by professionals' donation under permission of patients will be used, of this, 50 will be employed to pilot tests and other 150 will be divided in ten groups (n=15), in agreement with the technique of instrumentation and employed filling:G1 - ProTaper system and the single cone with instrument of conicity 25.08 in the length of work (CT), G2 - ProTaper system and System B Obtura II with instrument of conicity 25.08 in the length of work (CT), G3 - ProTaper system and Thermafil with instrument of conicity 25.08 in the length of work (CT), G4 - K3 system and the single cone with instrument 35.04 in the CT, G5 - K3 system and System B Obtura II with instrument 35.04 in the CT, G6 - K3 system and Thermafil with instrument 35.04 in the CT, G7 - ProTaper system and RealSeal with instrument 25.08 in the CT, G8 - K3 system and RealSeal with instrument 35.04 in the CT, G9 - ProTaper system and GuttaFlow with instrument 25.08 in the CT and G10 - K3 system and GuttaFlow with instrument 35.04 in the CT. After the filling, there will be carried out ten cross sections seriated in intervals of 0,5 mm. The sections obtained will be fotomicrografadas with help stereomicroscopy of increase of 40X. The images will be calibrated and analysed in the program Image Tool 3.0 ® for determination of the area from sealer, gutta-percha and empty spaces. The sections carried out to 2, 4 and 6 mm of the anatomical apices, will be analysed also under confocal microscopy with increase of 100x for determination of the penetration and adaptation from the sealer in the dentine radicular. For statistical analysis of the results the tests will be selectedf Kruskall-Wallis and Miller tests. (p <0,05). (AU)
