PCR molecular monitoring of hepatotoxic cyanobacteria in water supply systems at the metropolitan region of São Paulo-SP

Abstract

The cyanobacteria blooms in Brazilian aquatic ecosystems are increasingly more frequent, giving rise to several harmful consequences to both Public Health and the environment. Some cyanobacteria are able in producing microcystins, namely, hepatotoxins released to the water causing poisoning and promotes tumoring in mammalians. The presence of specific genes drives the production potentiality of microcystin. In this sense, toxic cyanobacteria are detected by means of genes molecular markers which code to the enzymes involved in the microcystin biosynthesis. According to the Portaria Ministério da Saúde 518, effective from April 2005 on, the companies in charge of water supply are compelled to provide systematic monitoring on the occurrence of cyanobacteria and microcystins in untreated water (from the source) , and in treated water intended for public supply. There are currently several kinds of analysis being used for that purpose, as e.g. bioassays with mice , ELISA and employing HPLC. However, those methodologies present some shortcomings and drawbacks concerning cost, task-speed, infrastructure, need of man-power training, etc. This project has as its main goal the validation of the molecular markers technique for microcystins (Method for the Detection of Microcystin is situ by means of Molecular Markers - PI0400576-7). The achieving of this goal will be accomplished by the use of samples obtained by technicians from SABESP in 6 reservoirs belonging to the most important water supply systems in the state of São Paulo metropolitan region. Microcystin analysis through imunoenzimatic (ELISA) and analytic (HPLC) methods will be compared with the obtained results. (AU)
