Purification and immobilization of the native microbial lipases for the enantioselective esterification of racemic ibuprofen


The present project plans to evaluate different purification and immobilization techniques of lipases obtained from Aspergillus niger and Aspergillus terreus with the purpose of developing processes that make possible the reutilization of enzymes as well as to obtain enzymes with better stability, activity and enantioselectivity for the resolution of (R,S)-ibuprofen. These microorganisms, isolated and selected in previous studies, have shown biocatalytic potential, considering their thermo-stability and good enantioselectivity for the resolution of this drug. The enantioselectivity will be evaluated from the preferential esterification reaction for one of the enantiomeric forms of the drug with n-propanol in medium containing isooctane, with and without the presence of ionic liquids. The kinetic and stability parameters will be compared before and after the purification and immobilization techniques. The experiments will be carried out with the use of chromatography (open column, CG and HPLC, using both normal and chiral columns) and the enantiomeric ratio (E) will be determined based on the conversion (c), enantiomeric excess of the substrate (ees) and of the product (eep) values.
