EMU Científico: Acquisition of a mass spectrometry platform for the advanced characterization of protein glycosylation and structure in healthy and disease conditions.


This proposal aims to implement a multiuser mass spectrometry-based platform specialized in glycobiology and structural proteomics by acquiring an Orbitrap Eclipse Tribrid coupled to a Vanquish Neo liquid chromatographic system. The technical features of this equipment such as different fragmentation techniques, ion mobility separation, MSn acquisition methods, data acquisition versatility, high accuracy, and resolution will improve the identification and quantification of glycoproteins, intact glycopeptides, and crosslinked peptides in complex biological systems. These analyses will aim at understanding the role of glycans and protein conformational changes in pathophysiological processes and their implication as biomarkers and therapeutic targets. The orbitrap Eclipse Tribrid will allow the implementation of analytical strategies for identifying and quantifying these biomolecules and acquiring high-quality data that will benefit postgraduate students, researchers, and companies by increasing international competitiveness. This equipment does not exist in Brazil, and its availability will help advance our glycobiology and structural proteomics knowledge from basic science to biotechnological and translational applications.
