Evaluation of cytotoxic, genotoxic, epigenetics, and antiangiogenic effects of the associated nutraceuticals diallyl disulfide and vitamin D in human colorectal adenocarcinoma cell lines

Abstract

The consumption of nutraceuticals is considered a strategy in cancer prevention, and the possible mechanisms of action include cell signaling pathways, cell cycle, modulation of redox balance, induction of DNA damage, epigenetic mechanisms and anti-angiogenesis. The data from the association between nutraceuticals showed that acting together can result in synergistic actions, with better responses in inhibiting tumor cell proliferation and inducing apoptosis. In this context, in vitro studies, in different cell culture systems, should present greater similarity with the in vivo conditions and the tumor microenvironment, and these models are currently recommended for pre-clinical assays. The present proposal intends to investigate at the molecular level if the association between the nutraceuticals diallyl disulfide (DADS) and vitamin D (VD) presents better responses in the induction of cell death, genotoxicity, modulation of gene expression, epigenetic alterations and anti-angiogenic effects in Caco-2 and HT-29 cells (human colorectal adenocarcinoma) in monoculture or 3D co-culture with HUVEC cells (human umbilical vein cells), than treatment with the nutraceuticals alone.
