Vibrational spectroscopic analysis of saliva: an innovative approach to real time investigation

Abstract

Biofluids hold great promise in terms of early diagnosis of various types of diseases, as well as therapeutic monitoring. In particular, saliva can be used for this purpose, and is easily accessible. Among the different methods of analysis to characterize biochemical dosage, including hemography, none can provide a simple disease and therapy associated profile in real time. In this regard, techniques such as High-Pressure Liquid Chromatography (HPLC) associated with mass spectrometry, although qualitatively important for biofluid analysis, are complex and require extensive preparation of the sample. In contrast, vibrational spectroscopy has the advantage of providing biochemical information from the samples evaluated in real time and without any preparation. Research using this technique is in its early phases, so that many steps to standardize the sampling protocols need to be performed. In this study, the interdisciplinary training of the researcher in both Dentistry and Analytical Sciences, as well as the experience of the international research field of biospectroscopy, will provide a unique opportunity to develop and standardize protocols for analysis of saliva, in the participating universities with the collaboration of three international Institutions (Ireland, Scotland and France). The aim of this study is to evaluate whether vibrational spectroscopic analysis of saliva is a useful technique to diagnosis Cancer in early stages, as our final aim. However, many optimization considerations should be addressed, including: method of collection, sample preparation, etc. This proposal will be divided into five sub-projects: 1) standardization of biofluid characterization in healthy subjects as a comparator group; 2) investigation of patient confounding factors (e.g. smokers vs non smokers); 3) potentially malignant conditions; 4) Oral Cancer and healthy donors normal. The project will be carried out over a time period of 4 years and engage professionals and academics from different fields, including Dentistry, Medicine, Biology, Biomedical Engineering, Chemistry, Physics and Computer Science. It will be performed at the University of Taubate, Centro Universitário Braz Cubas, in collaboration with, Clínicas de Estomatologia da Equipe CK Estomatologia, Universidade Federal do Estado de São Paulo, Technological University Dublin, University of François - Rebelais Tour and University of Strathclyde.
