Nanometric liposomes coated with resistant type 3 starch as novel delivery for proviotics: preparation, characterization and release behavior

Abstract

The demand for probiotics grown up due to the increased consumption of functional foods that, in addition to providing fundamental nutrition, have beneficial health impacts. Although there are several products on the market with the addition of probiotics, it is known that these bacteria are sensitive to various factors such as pH and oxygen and may not remain viable during the shelf life of a product and when passing through the human gastrointestinal tract. In developing this research, we seek to obtain an efficient nanoencapsulated functional compound, resistant to changes in pH and the human digestive system, and thereby achieve a high economic potential. In this context, the objective of this work is the development of nanoliposomes for application as probiotic delivery systems, in order to achieve a prolonged effect after their application. For this, resistant starch type 3 in different concentrations (0.5, 1.0 and 1.5 mg / mL) will be incorporated into the liposome interface, obtained from root starch, contributing to the modulated release of the encapsulated functional compounds. The nanostructures obtained will be characterized in terms of their physical-chemical properties, encapsulation efficiency and performance in the protection of encapsulated compounds. To evaluate the applicability of liposomes, tests will be carried out with free and encapsulated probiotic cells, produced with and without a type 3 resistant starch coating, exposed to controlled environmental conditions that resemble those of in vivo experiments, to evaluate their survival rates, using an in vitro model (SHIME). When using coated and uncoated liposomes, it is intended to assess whether the addition of resistant starch will promote an increase in the life and viability of probiotics during storage and in the gastrointestinal tract, as well as promoting their controlled release. It is expected, with the results of this research, that the new product generated will become responsible for 50% of Starch Tech Innovations' turnover, aiming at future operations in the probiotics market in Brazil, expanding our areas of operation. The global probiotic market has shown an average annual growth of 7.6%. Latin America is the region with the highest consumption of probiotic products in the world, with Brazil representing 52% of the market with a growth trend of around 11% until 2022. (AU)
