Diversity of piroplasmids in wild birds and mammals from Brazil

Abstract

Piroplasmids are tick-borne apicomplexan protozoa that comprise Babesia, Rangelia, Cytauxzoon and Theileria genera. There is a wide diversity of species that affect different taxa of mammals and birds, which can cause diseases in animals and humans. Studies aiming to detect piroplasmids in the Brazilian fauna, which is extremely diverse, show that different and new species of these protozoa circulate in the country. The present study aims to investigate the occurrence and determine the genetic diversity of piroplasmids in different taxonomic groups of vertebrates in Brazil. For this purpose, DNA aliquots will be extracted from 2,494 biological samples from wild animals and associated ectoparasites (for some of them) belonging to several taxonomic groups (327 Pilosa, 27 Cingulata, 878 Chiroptera, 288 Artiodactyla, 219 Carnivora, 129 Rodentia, oito Didelphimorphia, 118 Perissodactyla e 500 Passeriformes) from different states all over Brazil. The biological samples will be submitted to PCR assays based on the 18S rRNA, hsp-70, cox-1, cox-3, cytB, B-tubulin genes, and ITS-1 and ITS-2 intergenic regions. Positive samples will be purified and sequenced by the Sanger method. Samples with multiple peaks in the chromatograms will be subjected to gene cloning. The phylogenetic positioning of the obtained sequences will be performed by dendogram construction. Finally, the alignment of the sequences will be used to calculate the diversity of nucleotides and the level of polymorphisms for the analysis of the genetic diversity of the agents. The genealogy of the nucleotide sequences will be performed using the Splitstree program. The present study will shed light on the diversity of piroplasmids among Brazilian wild fauna. (AU)
