Effects of caffeine exposure on mitophagic pathways: actions over myogenic progenitor cells differentiation

Abstract

The skeletal muscle regenerative process is critical for tissue functional recovery after any lesion stimuli, micro-injuries or large injuries. Thus, the ability to regenerate allows the skeletal muscle to keep the homeostasis and, consequently, the whole body's health. In our previous study (under revision), we have demonstrated that in Parkin's absence, myogenic progenitor cells present a reduced autophagic process with delayed skeletal muscle regeneration. The caffeine has been associated with the prevention of neurotoxicity related to Parkinson. However, few studies have approached the caffeine effects over myogenic progenitor cells. Our hypothesis is caffeine could act as a therapeutic agent by regulating the mitochondrial population since it has been demonstrated that caffeine can induce autophagy in several cell types and mitochondrial biogenesis in muscle fibers. Thus, our main objective is to investigate the effects of caffeine over the differentiation process of myogenic cells in vitro, and over skeletal muscle regeneration on wild type (WT) and Parkin knockout (Parkin-/-) animals. To improve our knowledge about caffeine metabolism, we also aim to verify whether Parkin-/- present a deficient capacity to absolve and to metabolize caffeine, as suggested to humans with Park2 mutations. That information will be relevant to indicate new ways of caffeine action on skeletal muscle tissue, which can be used further to myopathies treatment. (AU)
