Flame retardants have effectively reduced the deaths and injuries caused by fires, and thus the need for their use in various consumer goods is indisputable. Among these compounds, polybrominated biphenyls (PBDEs) congeners are very efficient in mitigating the combustion process, reducing the spread of flames, besides being low cost. However, several studies have demonstrated the ability to induce various toxic effects after exposure to these compounds. Thus, there is an international tendency to replace these compounds, and aluminum diethylphosphinate (ALPI) has been suggested as a potential alternative retardant. However, in order to safely replace considering general population and environment, the toxicity of this new compound is necessary to compare its safety with compounds that are already available in the market. Within this context, the present project aims to evaluate the induction of early neurobehavioral effects induced by ALPI in comparison to the effects induced by selected PBDEs. Therefore, we propose the establishment of a strategy of multi-biomarkers using initial stages of Danio rerio, aiming to estimate CNS effects. Thus, we propose the combined use of two high-performance tools: i) the Zebrabox equipment, which video monitors the swimming behavior of larvae in different light conditions; and ii) the software, DanioScope, which evaluates the profile of embryonic movement. To complement these analyzes, we propose the evaluation of the activity of the cholinesterase enzyme, as a biochemical biomarker of neurotoxicity. In addition, since the maintenance of homeostasis of organisms against chemical stressors can be energetically costly, we also propose the use of biomarkers of energy balance. In this way, the present project aims to realize an integrated approach of the neurotoxicity induced by representatives of flame retardants, providing subsidies for decision making regarding the substitution of the currently used compounds. In addition, the results will be useful in establishing safety standards for these compounds, with a view to human health and the conservation of aquatic communities. Still, the approval of this proposal will provide a great development in the research line of the Laboratory of Environmental Toxicology, in addition to complement the tests already carried out. The project will also contribute greatly to the training of specialized human resources, since it will count on the participation of masters and doctoral students, as well as post-doctoral students. Finally, the project will increase collaboration between researchers in the short and medium term, because the biomarkers strategy could be used in other studies of effects of compounds in the CNS, both in Toxicology and Pharmacology.
