Study of the effects of progesterone and the hypercaloric diet on the interactions between pancreatic beta cell and adipose tissue


Obesity has been taking on alarming dimensions at the global level, which reflects changes in dietary and nutritional patterns. Associated with the pathogenesis of this condition is the increased peripheral resistance to insulin, which may be related to the increased risk that obese women have to develop gestational diabetes. Pregnancy, per se, is a condition where physiological resistance to insulin isobserved and progesterone is one of the hormones responsible for this condition.Considering this scenario, here we intend to investigate the effects of progesteroneand the hypercaloric diet on the interactions between pancreatic beta cell and adipose tissue. Two approaches will be used. In vivo, female C57B1 mice fed standard or hypercaloric diet will be used, and followed up general and metabolic parameters, such as: body weight, energy expenditure, fasting glucose, insulin and glucose tolerance tests. Immunohistochemical analyzes of adipose tissue and pancreas, study of insulin secretion by isolated islets and plasma adiponectindetermination will also be performed. In the in vitro approach, beta cell lines, RINm5Fand INS-1E, will be used to analyze the combined effects of fatty acids and steroidhormones on cell parameters such as: cell membrane integrity, percentage of cellswith fragmented DNA, cell proliferation and analysis of the production of reactivespecies. Thus, we hope to contribute with data on the effects of progesterone on thecommunication between beta cells and adipose tissue, so that at the end of this workwe will understand a little more about the relationship between obesity and DG.
