Development of a fluoride varnish formulation containing nano-sized sodium trimetaphosphate: effects on enamel erosion and on erosive enamel wear using in vitro and in situ protocols

Abstract

The aim of the present study is to assess the effect of fluoride varnishes supplemented with nano-sized sodium trimetaphosphate (TMP) on enamel erosion and on enamel erosive wear, using in vitro and in situ protocols. For this purpose, the study will be divided into 3 subprojects, in which blocks of bovine dental enamel will be used as substrate. The varnishes to be tested are: (A) Placebo (without NaF or TMP), (B) 5% NaF, (C) 5% NaF + 5% micrometric TMP, (D) 5% NaF + 5% nano-sized TMP, and (E) 5% NaF + 2.5% nano-sized TMP, which will be applied only once on the blocks. Subproject 1 (S1) will assess the effect of varnishes on initial erosive lesions, both on surface hardness (SH) recovery and on the protection against SH loss under a further erosive challenge in vitro. Subproject 2 (S2) will assess the effect of the same varnishes on enamel erosive wear resulting from exposure to citric acid, followed or not by abrasion by toothbrushing with a fluoride-free dentifrice in vitro. In Subproject 3 (S3), varnishes A, B and C, as well as varnish D or E (to be determined from the results of S2) will be tested using an in situ/ex vivo protocol, in individuals brushing with a fluoridated dentifrice. The number of blocks (S1 and S2) and subjects (S3) will be determined in pilot studies. Blocks will be evaluated by SH (S1, S2 and S3), profilometry (S2 and S3), cross-sectional microhardness (S2 and S3) and computed microtomography (S3). The homogeneity and homoscedasticity of the data will be determined prior to the statistical analysis. If the use of parametric tests is possible, the data will be submitted to ANOVA (1- and 2-way), followed by the Student-Newman-Keuls test. Otherwise, the corresponding non-parametric tests will be applied. The level of significance adopted will be 5%. (AU)
