Proteomic profile of bacteria from root canals of symptomatic or asymptomatic teeth with apical periodontitis

Abstract

Endodontic infections are caused by a multi-species community of bacteria. It is difficult to infer the physiology, function and microbial pathogenicity with methods of microbiological identification and quantification of endotoxins. Thus, proteomic analysis is a technique that can revolutionize the study of the pathogenesis of endodontic infections. The present study aims to: analyze the proteomic profile of bacteria present in root canals of teeth with symptomatic and asymptomatic apical periodontitis; to broaden the understanding of the role of the proteome in the pathogenesis of periapical diseases; to investigate the correlation of the composition of the bacterial proteins detected with the presence of clinical signs and symptoms. Samples will be collected from 18 patients referred for root canal treatment or emergency treatment at the Araçatuba Dental School FOA - UNESP. Teeth with primary endodontic infection, symptomatic (n=9) or asymptomatic (n=9) and presence of visible periapical lesion on the radiograph will be included. The identification of the peptides will be done in a nanoACQUITY UPLC-Xevo QTof MS system (Waters), protein identification will be obtained using ProteinLynx Global Server software (PLGS) version 3.0 using the National Center's bacterial protein databases of Biotechnology Information. The data will be analyzed statistically with SPSS for Windows (SPSS Inc., Chicago, IL) (p <0.05). (AU)
