GENETIC DIVERSITY AS A STRATEGY FOR THE MANAGEMENT AND USE OF GERMPLASM OF Vitis sp. OF THE AGRONOMIC INSTITUTE

Abstract

In most fruit plants the conservation of germplasm banks is ex situ in the field, due being perennial and highly heterozygous. The management of such banks becomes complex, especially when many accesses are involved. Redundancy should be kept to a minimum and homogeneity and reliability in the material should be ensured, while the introduction of new accesses should be optimized. Thus, the identification of plant material is therefore crucial and represents the first step in the management of germplasm. The project aims to analyze the genetic diversity and kinship of grapevine accesses from the Germplasm Active Bank of the Instituto Agronômico, for this purpose, accesses will be genotyped with microsatellite markers. The genetic data obtained will be analyzed to determine the genetic structure of the populations, to quantify the allelic diversity and to suggest the composition of a core collection to capture the maximum genetic diversity within a minimum sample size. Molecular analysis will allow a better understanding of the level and structuring of available diversity. This will guide the accomplishment of the crosses in the scope of the grapevine breeding program of the institution. The development of a core collection will contribute to reducing the number of duplicates, minimizing maintenance costs in the field and becoming more representative for research use. Additionally, this study aims to generate a molecular database that will facilitate the management of Vitis germplasm and its use for later genetic and genomic breeding programs. Through this study of diversity, we intend to establish a fingerprint (genetic identities) for molecular characterization and identification of cultivars from the breeding program of IAC already registered in MAPA and future new cultivars from crosses, a criterion due to the future requirement of this organ for registration in the National Register of Cultivars of Vine (RNC). It is also intended to identify the probable parents of the hybrid SR 501-17, coming from the vine improvement program of the IAC. (AU)
