Abstract
This study will investigate the action of the association between systemic medication (oral bisphosphonate) an several medicines that will be locally applied in the surface of implants that will be installed in rat' maxilla. The medicines that will be used this study in order to functionalism the implant's surface will be Terpy (nitric oxide donator), teriparatide and genisteine. This project will be executed in two steps. In the first step, it will be performed the characterization of the implant' surface after the medicine' functionalization. After the validation with the tests that will be executed (physical, chemical and biological tests), the second step will be performed with the in vivo experiments. For the second step, Wistar rats, female, adults will be submitted to bilateral ovariectomy and they will receive hyper caloric diet, mimetizing metabolic syndrome condition. Simultaneously, Wistar rats, female, adults will be submitted to sham surgery and they will received balanced diet and water ad libitum. Animals will expect for 30 days and then, risedronate treatment will be started (0,70mg/kg), gavage, once a week. Another group of animals that will be submitted to ovariectomy surgery and that will receive hiperlipidic diet (metabolic syndrome) will receive vehicle instead of risedronate. After 30 days, all of the animals will be submitted to upper molar extraction, bilateral. After 30 days, the same animals will receive implants that will be installed in the region where the tooth extraction surgeries were performed. Implants will have the surface funcionalized with the medicines or will have no surface treatment, according to the experimental group that was proposed in this study. The euthanasia of the animals will be performed at 60 days after the implant's installation and the right side maxila will be reserved for biomechanics analysis and RT PCR . The left side maxilas will be reserved for the calcified analysis, through fluorochromes injection (calcein and alizarin) and computadorized microtomography analysis. The same samples will be reserved to receive Alizarin red and Steven el blue staining in order to be submitted to histometrical analysis (BIC and neoformed area). Quantitative data will be submitted to homocedasticity test in order to select the appropriate statistical test. (AU)
