Hydroxyapatite/carbon nanotubes nanocomposite as reinforcement in fibrin sealant derived from snake venom for bone regeneration


Fibrin sealants, developed from human hematological derivatives mimic the physiological clots, being widely used in general and cosmetic surgeries and in postoperative for their hemostatic and adhesive action. Fibrin sealant developed from the venom of Crotalus durissus terrificus (rattlesnake) is a heterologous derivative like thrombin that converts fibrinogen to fibrin. It is a natural, gel-like, biodegradable, bioabsorbent, non-toxic and non-immunogenic product. However, the use of this sealant as a matrix for bone regeneration has never been studied. Hydroxyapatite/carbon nanotubes nanocomposites produced by the chemical synthesis assisted by the ultrasound technique are bioactivity, non-cytotoxic and present bactericidal effect. The aim of this project is to evaluate the use of hydroxyapatite/carbon nanotubes nanocomposites as reinforcement in fibrin sealant derived from snake venom for bone regeneration. In vitro and in vivo experiments will be performed. In vitro experiments will consist of evaluation of cell adhesion and proliferation using culture of human osteoblast cells. In vivo experiments will consist of evaluation of the histological coverslips of bone defects in Rattus norvegicus albinus rats; Wistar. These coverslips will also be characterized by Raman and infrared spectroscopy to monitor the formation of the osseointegration process of the implanted samples. The project will be forwarded to the Ethical Committee on Animal Use of Universidade Brasil. All data will be analyzed by means of the normality test, and the parametric results were submitted to the two-way ANOVA (time versus samples) test complemented by the Tukey post-test to verify the statistical differences between groups. It is expected that the hydroxyapatite/carbon nanotubes nanocomposites assist in the process of osseointegration, thus reducing their time. (AU)
