Abstract

Anurans have pigment cells (melanocytes and melanomacrophages) in different organs and membranes, which have melanin. The visceral pigmentation can be related to residual melanin accumulation, may have a role in the immune system, antioxidant functions, and protect the tissue. One of the objectives of this study is to describe the morphometric characteristics of the hepatic melanomacrophages (MMs) and characterize the liver histology in ten species of anurans. Data analysis includes morphological and stereological analyses, quantification of melanin, hemosiderin and lipofuscin. The other purpose, according to the hypothesis that the Benzo[alpha]pyrene (BaP) is a hepatotoxic contaminant will be: to evaluate the dispersion of the melanin area of MMs (quantify the melanin production); to evaluate phagocytic activity of MMs (BaP changes in the cytoskeleton); to evaluate genotoxic effects. For the histological analyzes the procedures for inclusion in historesin and light microscopy analysis of the area of melanin and phagocytosis will be followed. The amount of melanin will be measured by Elisa. For microtubules, pellets with MMs will be analyzed (by fluorescence microscope). For nuclear abnormalities, the blood will be extracted and the procedures for analysis will be followed. These morphometric characteristics of liver tissue and properties of the MMs can be used as functional biomarkers for the assessment of anurans' health and to test the functions of visceral pigmentation related to chemical pollutant factors.
