Abstract

Recent efforts in periodontal treatment have focused on regenerative approaches based in cells. The membranes have gained full attention on the repair/ regeneration of periodontal defects due to their ability to allow osteoblastic proliferation and to support the mineralized matrix formation. The obtainment of the cell sheets through the detachment of the cell layer from the culture dish (without their dissociation) is a method that has been highlighted in the current scenario. Additionally, the periodontal ligament stem cells (PDLSCs) are able to differentiate in cementoblasts/ osteoblasts, adipocytes and collagen forming cells, contributing in the periodontal tissue regeneration. It is known that the protease activated receptor type 1 (PAR1) has a crucial role on the initiation and maintenance of inflammation and fractures repair, by regulating several aspects of the osteoblastic functions. Recently, our group has observed that PAR1 activation is able to increase osteogenic activity of PDLSCs. However, the mechanism by which this process is accomplished is not completed elucidated and its role through the use of cell sheets has not been studied yet. Therefore, the main objective of the present work will be to evaluate the effect of PAR1 activation on the osteogenic activity in cell sheets obtained from PDLSCs. Thus, we will evaluate the biologic in vitro and in vivo efficacy of cell sheets activated by the PAR1 activating peptide in order to develp a new therapeutic approach for periodontal defects. (AU)
